Departamento de Biología Celular e Histología, 2012) Santi, R.; Franchi, A.; Villari, D.; Paglierani, M.; Pepi, M.; Danielli, D.; Nicita, G.; Nesi, G.Tumours of perivascular epithelioid cells (PEComas) are a heterogeneous group of uncommon mesenchymal neoplasms which exhibit a peculiar immunohistochemical co-expression of muscle and melanocytic markers. PEComas occur at various visceral and soft tissue sites, generally with a benign clinical course. Nevertheless, there has been evidence of cases having an unfavourable outcome, thus prompting investigation of pathological criteria for malignancy. A sclerosing variant of PEComa, more frequently encountered in the retroperitoneum of middle-aged women, has been reported. Prognosis has generally been regarded as favourable and complete surgical excision appears to be adequate treatment. To the best of our knowledge, only two cases of sclerosing PEComa displayed high-grade malignant morphology and were associated with adverse outcome. An additional case of retroperitoneal sclerosing PEComa with a two-year follow-up and indolent behaviour is herein described. Light and electron microscopy were performed, along with immunohistochemical analysis. Further studies are needed to clarify the histogenesis and to predict the biological behaviour of this uncommon entity.

Departamento de Biología Celular e Histología, 2012) Maounis, Nicoletta F.; Dráberová, Eduarda; Mahera, Heleni; Chorti, María; Caracciolo, Valentina; Sulimenko, Tetyana; Riga, Dimitra; Trakas, Nikolaos; Emmanouilidou, Aphrodite; Giordano, Antonio; Dráber, Pavel; Katsetos, Christos D.We and others have previously shown that increased expression and altered compartmentalization of γ-tubulin may contribute to tumorigenesis and tumor progression (J. Cell Physiol. 2009;223:519-529; Cancer Biol. Ther. 2010;9:66-76). Here we have determined by immunohistochemistry the localization and cellular distribution of γ-tubulin in clinical tissue samples from 109 non-small cell lung cancer (NSCLC) cases. The expression and distribution of γ-tubulin protein and transcripts was also determined in the NSCLC tumor cell lines NCI-H460 (HTB-177) and NCI-H69 (HTB-119) by immunocytochemistry, quantitative immunoblotting and reverse transcription quantitative real-time PCR (RT-qPCR). Polyclonal and monoclonal anti-peptide antibodies recognizing epitopes in the C- or N-terminal domains of γ-tubulins and human gene-specific primers for γ-tubulins 1 (TUBG1) and 2 (TUBG2) were used. In non-neoplastic cells of the airway epithelium in situ, γ-tubulin exhibited predominantly apical surface and pericentriolar localizations. In contrast, markedly increased, albeit heterogeneous and variously prominent γ-tubulin immunoreactivity was detected in clinical tumor specimens and in the NCI-H460 and NCI-H69 cell lines, where tumor cells exhibited overlapping multi-punctate and diffuse patterns of localization. Co-expression of γ-tubulin and Ki-67 (MIB-1) was detected in a population of proliferating tumor cells. A statistically significant increase of γ-tubulin expression was found in Stage III compared to lesser stage tumors (p<0.001 v. Stages I/II) regardless of histological subtype or grade. By quantitative immunoblotting NCI-H460 and NCI-H69 cells expressed higher levels of γ-tubulin protein compared to small airway epithelial cells (SAEC). In both tumor cell lines increase in TUBG1 and TUBG2 transcripts was detected by RT-qPCR. Our results reveal for the first time an increased expression of γ-tubulin in lung cancer

Departamento de Biología Celular e Histología, 2012) Abreu, Camila C.; Nakayama, Priscilla A.; Nogueira, Clayton I.; Mesquita, Leonardo P.; Lopes, Priscila F.R.; Varaschin, Mary S.; Seixas, Josilene do N.; Ferreira, Enio; Bezerra Jr, Pedro S.The present study describes the use of a microwave processing protocol for the rapid histopathological and immunohistochemical diagnosis of bovine rabies. Immunohistochemistry has been used for rabies diagnosis in formalin-fixed tissue with satisfactory results, although the time to diagnosis is considerably longer than that with direct immunofluorescence. The protocol provided a provisory histopathological rabies diagnosis in approximately three and half hours and the immunohistochemical diagnosis was available after six hours. The protocol achieved 100% correlation with direct immunofluorescence and is a promising method, particularly in situations in which only material in formalin is available for diagnosis or when the refrigeration or transportation of biological material is difficult.

This study aims to explore whether the protective effect of SO2 on ISO-induced myocardial injury was mediated by the restoration of calcium homeostasis disturbance in cardiomyocyte. Methods and results: Rats were randomly divided into four groups: ISO group, ISO+SO2 group, control group and SO2 group. Content of Ca2+ in H9c2 cells was assayed using confocal microscope, and cardiac function parameters were measured by echocardiography. Plasma biochemical values and myocardial ultra-structure changes were measured. Meanwhile, the activity, protein and gene levels of sarcoplasmic reticulum Ca2+ ATPase (SERCA), and protein and phosphorylation of phospholamban (PLN) were detected. We found SO2 derivatives could restore the decreased cardiac function, the abnormal lactate dehydrogenase, creatine kinase, alpha-hydroxybutyrate dehydrogenase, potassium, calcium, blood urea nitrogen and the damaged myocardial ultra-structure in rats, and regulate the increased Ca2+ content in H9c2 induced by ISO. In addition, compared with ISO group, the decreased activities, protein and mRNA level of SERCA, as well as the decreased protein phosphorylation level of PLN in myocardial tissues were increased in ISO+SO2 group. Conclusion: SO2 derivatives might relieve calcium overload in association with the upregulating expression of SERCA and p-PLN/PLN by myocardial tissues in rats with ISO-induced myocardial injury

Departamento de Biología Celular e Histología, 2012) Pivko, J.; Makarevich, A.V.; Kubovičova, E.; Ostro, A.; Hegedušová, Z.; Louda, F.The aim of the study was to define interrelationships between histopathological alterations in ovarian antral follicles and body condition in dairy cows with a tendency to emaciation (BCS 1 and 2) compared with dairy cows with normal body condition (BCS 3). The ovaries were recovered from slaughtered cyclic dairy cows (at the luteal phase of the cycle) of Czech Fleckvieh and Holstein breeds at different times of the post-partum period. The animals were estimated as belonging to certain grade of body condition score (BCS) according to a 5-point scale. Only dairy cows with BCS1 (emaciation; n=6), BCS2 (tendency to emaciation; n=5) and BCS3 (optimal body condition status; n=6) were available for the experiment. The ovarian samples were embedded into Technovit 7100 resin; the tissue sections were stained with buffered basic fuchsine with toluidine blue. For acidic mucopolysaccharides (aMPS) a combination of PAS-technique with Alcian blue was used. Histological analysis showed that emaciation was associated with an increased occurrence of late (cystic) and luteinization-related atresia in granulosa and theca cells and increased levels of aMPS in small atretic follicles. Our observations indicate that dairy cows with a tendency to emaciation (BCS 2) or emaciated (BCS 1) have elevated occurrence of late atresia and atresia with luteinization, while initial atresia is less. This expands our basic knowledge of ovarian histopathology providing new insight into the association of antral follicle atresia and body condition status in dairy cows
