    Evaluation of a commercial double-recognition ELISA for caprine tuberculosis diagnosis: sensitivity, specificity, and correlation to macroscopic and microscopic lesions

    Ante-mortem diagnosis of caprine tuberculosis, as in other ruminant species, is a complex process. Therefore, it is sometimes necessary to apply a combination of methods that increases the sensitivity of diagnosis while maintaining adequate specificity. In the present study, we analysed the use of ELISA based on a double-recognition methodology for the diagnosis of goat tuberculosis. Serum samples from animals with tuberculosis, confirmed by histopathological studies and presence of Ziehl-Neelsen-positive bacteria in the lesions, were used to assess sensitivity. Sera from flocks without tuberculosis were used to assess diagnostic specificity. The relationship between ELISA results and the type of tuberculous lesions, both macroscopic and microscopic, was analysed. The sensitivity and specificity of ELISA test were 69.9% and 97.9%, respectively. Sensitivity was higher in animals with macroscopic lesions, indicating more advanced forms of the disease. Similarly, a higher sensitivity was found in animals with microscopic lesions associated with active tuberculosis, such as cavitary lesions.

    Value of anatomopathological examination in goats with a positive comparative intradermal tuberculin test as part of a tuberculosis control programme

    Background: Caprine tuberculosis is a chronic infectious disease caused by Mycobacterium bovis and Mycobacterium caprae. A tuberculosis control programme has been implemented using the comparative intradermal tuberculin (CIT) test. However, infection with Corynebacterium pseudotuberculsis and infection with or vaccination against Mycobacterium avium subspecies paratuberculosis (MAP)may have a negative impact on specificity. Therefore, a postmortem examination is necessary to verify the tuberculosis status of each animal where positive predictive value is low. Methods: Thirty-seven CIT-positive goats from 14 herds with a previously tuberculosis-free status were slaughtered between 2015 and 2020, and postmortem examination was carried out at the School of Veterinary Medicine of Murcia. Results: Anatomopathological examination ruled out the presence of tuberculosis lesions. In 23 of the goats, the anatomopathological examination revealed the presence of the paucibacillary form of paratuberculosis (associated with MAP infection), and in another six goats, Corynebacterium pseudotuberculosis infection was confirmed. Positive reactors have not reappeared in any of the herds in the annual CIT tests up to the year 2023. Limitations: Only those herds that sent their positive animals to the veterinary school were included in the study. Participation in the postmortem analysis was notmandatory for farmers. Conclusions: Anatomopathological examination is a useful, rapid and economically viable tool to determine the tuberculosis status of goat herds.

    Prevalence of Mycobacterium avium Subsp. paratuberculosis in feral pigeons (Columba livia) associated with difficulties controlling Paratuberculosis in a bovine herd (Fighting Bull Breed)

    A bovine herd with a high prevalence of paratuberculosis (PTB) cohabiting with a population of pigeons was studied (2011–2020). After finding the disease in 2011, annual monitoring was performed in 2012–2014 by obtaining blood samples for ELISA and intradermal tuberculinization (IT) tests for Mycobacterium avium subsp. paratuberculosis (MAP). Positive animals were eliminated. PTB prevalence dropped from 10% to 0% but returned to similar values (9.5%) after 6 years without tests. In all animals, Ac values according to the optical density (OD) determined by ELISA increased each year and could be used to isolate herds close to the cutoff point to improve PTB control. Possible reservoirs were considered after evaluating the little success of the PTB control program, and the population of feral pigeons was studied. Specifically, 10% of the pigeon population (n = 13) was necropsied. Samples of intestine, feces, and foot skin for PCR study for MAP and samples of terminal intestine for histopathological analysis were taken. Eleven pigeons were PCR-positive against MAP, in the intestine (10/11), foot skin (3/11), and feces (1/11). The presence of MAP in pigeon feet could demonstrate its role as a mechanical disseminator of PTB, while the presence in pigeon intestine and feces could also suggest its role as a reservoir.
