    Morphologic changes and methodological issues in the rabbit experimental model for diaphragmatic hernia

    Summary. Fetal lung development may be impaired by some congenital anomalies or in utero events. Animal models are used to understand the pathophysiology of these diseases and explore therapeutic strategies. Our group has an interest in the prenatal management of congenital diaphragmatic hernia (CDH). Isolated CDH remains associated with a 30% mortality because of lung hypoplasia and pulmonary hypertension. On day 23 of gestation (pseudoglandular stage) CDH was created in both ovarian-end fetuses (n=28) in 14 time-mated pregnant white rabbits (hybrid of Dendermonde and New-Zealand White). At term (day 30) all survived operated fetuses and size-matched controls were harvested. Fetuses/lungs were assigned randomly to formalin fixation either under pressure of 25 cm H2O (CDH25 n=5; CTR25 n=5) or without (0 cm H2O (CDH0 n=7; CTR0 n=7). Fetuses and lungs were first weighed, and then the lungs were processed for morphometry. Pulmonary development was evaluated by lung-to-body weight ratio (LBWR) and airway and vascular morphometry. Surgical induction of CDH does reduce the LBWR to hypoplastic levels. The contralateral lung weight is 81% of what is expected, whereas the ipsilateral lung is only 46% of the normal. This was accompagnied by a loss of conducting airway generations, precisely, terminal bronchioles (TB), which were surrounded by less alveoli. The ipsilateral CDH lung demonstrated a thickened media in the peripheral arteries as well. As a result, in the severely hypoplastic ipsilateral lung, an airway fixation pressure of 25 H2O has no significant effect on the morphometric indices. The contralateral lung has a normal amount of alveoli around a single TB, which also behave like alveoli of the normal lung, i.e. expand under pressure fixation. The present study on severely hypoplastic lungs that never respirated, shows that in contrast to normal lungs, the morphometric indices are not significantly influenced by a difference in fixation pressure. Increasing fixation pressure seems to expand the lung only when sufficient alveolated parenchyma is present.

    Perfil epidemiológico de sífilis congénita en una microrregión en el interior del estado de Bahia (2007-2017)

    Objetivo:Analizar el perfil epidemiológico de Sífilis Congénita en 18 municipios en el interior del estado de Bahía entre 2007 y 2017.Método:Estudio descriptivo, transversal y cuantitativo de los casos confirmados de Sífilis Congénita en la microrregiónregistrados en el Sistema de Información de Agravios y Notificación.Resultados:En el período estudiado se confirmaron 39 casos, donde el 30,8% de las madres tiene entre 20 a 24 años, el 59% son pardas, el 46,2% tienen enseñanza fundamental incompleta, el 33,3% amas de casa, el 74,4% en el momento del parto / curetaje, el 41% recibió tratamiento inadecuado y el 38,5% de los socios no fueron tratados. Con respecto a los recién nacidos, el 69,2% fue diagnosticado con 0 días de vida, 74,4% con Sífilis Congénita reciente y 46,2% asintomáticos.Conclusión:Los datos revelan un serio problema de salud pública en la microrregión analizada apuntando también fallas en el sistema de notificación, observadas en el elevado índice de ignorados en las variables estudiadas.

    Prenatal and/or postnatal high-fat diet alters testicular parameters in adult Wistar Albino rats

    Here, we evaluated the effects of a high-fat diet during the prenatal and/or postnatal period on the metabolic parameters and testes of 4-month-old Wistar rats. The experimental groups, composed of male Wistar rats, were: C/C (n=8), HF/HF (n=8) (pups with the same diet as their dams, after weaning), C/HF (n=8), and HF/C (n=9) (pups with a different diet after weaning, from that of their dams). The biometric parameters, blood glucose levels, serum levels, the gonadosomatic index, sperm parameters, testes, and genital fat deposits were evaluated. The HDL-c serum levels were significantly lower in the C/HF group (P=0.0100), whereas animals in the HF/C group presented hypertriglyceridemia (P=0.0005). The sperm concentration was lower in the HF/HF group than in the HF/C group (P=0.0072), and sperm viability was lower in all groups receiving a high-fat diet (P<0.0001). The tubular compartment was the smallest in the HF/HF group (P<0.0001). The diameter of the seminiferous tubule was the widest in the HF/C group (P<0.0001). The height of the seminiferous epithelium in all groups was lower than that in the C/C group (P<0.0001). Testosterone and steroidogenic acute regulatory protein (STAR) expression levels were lower in the C/HF group (P=0.0218 and P=0.0215, respectively). The HF diet, regardless of the administration period, induced a limited number of metabolic changes, and modifications in the histoarchitecture of the testes and sperm parameters. These data suggest that a HF diet may cause disturbances in spermatogenesis and fertility impairment in adulthood.
