- PublicationOpen AccessEfecto de las secreciones de organoides de endometrio humano sobre el cultivo in vitro de embriones bovinos(Facultad de Veterinaria y el Servicio de Publicaciones de la Universidad de Murcia, 2023) Masegosa Domínguez, Alicia María; Navarro-Serna, Sergio; Pérez-García, Vicente; Coy, Pilar; Juan, Ángela; Romero Aguirregomezcorta, JonDurante los últimos 50 años, la fertilidad de las vacas lecheras de alta producción ha disminuido debido a la intensidad de la selección genética. Para superar este obstáculo, se ha recurrido a las técnicas de reproducción asistida. Sin embargo, la producción de embriones in vitro es un proceso relativamente ineficiente, con tasas máximas de blastocistos obtenidos de alrededor del 35%. Para solventar este problema, se planteó la hipótesis de añadir al medio de cultivo secreciones de organoides de endometrio, así, los embriones producidos in vitroestarían expuestos a un ambiente más fisiológico y el rendimiento de la técnica aumentaría. Para ello, se llevó a cabo la recolección de ovocitos bovinos procedentes de un matadero local, y se procedió a su maduración in vitro, fecundación in vitro y cultivo embrionario in vitro. El medio de cultivo embrionario se suplementó al 1% (v/v) con i) secreciones de organoides de endometrio humano (ya que no existe disponibilidad de secreciones bovinas), ii) medio de cultivo Boretto, y ii) gotas de matrigel con medio de cultivo Boretto, como controles de procedimiento. Además, se incluyó un grupo control sin ningún suplemento. Se evaluó el desarrollo embriona-rio a los días 2, 7 y 8 de cultivo. El porcentaje de blastocistos a día 7 y a día 8 post inseminación fue significa-tivamente mayor (P<0.05) cuando se añadió el medio de cultivo Boretto. Además, este tratamiento incrementó la cinética de desarrollo de los embriones a día 7 post inseminación, y la calidad de los blastocistos evaluada por el número total de células, con un 85.8% más que el control. Aunque las secreciones de endometrio humano no han dado el resultado esperado, sería necesario comprobar si las secreciones de organoides específicas de endometrio bovino, a distintas concentraciones, podrían incrementar el rendimiento de la producción in vitro de embriones bovinos.

Sin embargo, las investigaciones sobre el uso de estas técnicas en diversas especies muestran la aparición de anomalías en la placenta y en el cordón umbilical que puede estar relacionadas con la vascularización de embriones producidos in-vitro (IVP). De hecho, se sabe que el funcionamiento placentario y umbilical afecta al crecimiento fetal y el peso de las crías tanto en animales como en humanos, pero sus efectos a largo plazo son aún desconocidos. El objetivo de este estudio es analizar la morfometría vascular placentaria y umbilical de embriones porcinos producidos in-vivo por inseminación artificial (AI) e in-vitro añadiendo (RF-IVP) o no (C-IVP) fluidos reproductivos al medio de cultivo; y estudiar la relación entre los parámetros vasculares y la ganancia media diaria de los cerdos durante su crecimiento. Las muestras de placenta y cordón umbilical se recogieron después del nacimiento y fueron fijadas en formaldehído al 10 % para su inclusión en parafina. Se cortaron secciones histológicas de 5 μm de grosor que fueron teñidas con hematoxilina-eosina y fotografiadas a 5x para el análisis vascular y morfométrico con ImageJ® y Slide Viewer®. Los resultados de la morfometría vascular placentaria fueron similares entre los tres grupos. Sin embargo, el área vascular en los vasos de pequeño calibre (arteriolas-vénulas y vasos pequeños) fue significativamente más alto en el grupo C-IVP respecto de los otros dos grupos. En cuanto al cordón umbilical, el área (AI 43.1±12.8, IVP 56.6±14.9 mm2), perímetro (AI 26.4±3.9, IVP 30.5±4.7 mm), diámetro (AI 8.4±1.0, IVP 10.3±1.9 mm) y gelatina de Wharton (AI 36.9±12.0, IVP 48.9±12.8 mm2) fueron significativamente más altos en los embriones producidos in-vitro, mientras que los parámetros morfométricos arterial y venoso fueron similares entre los grupos (AI: 1.97±0.4 y 4.4±1.4 mm2, IVP: 2.5±0.6 y 5.6±3.0 mm2). Estos hallazgos demuestran que el crecimiento de los cerdos puede estar relacionado con el desarrollo vascular de la placenta y del cordón umbilical, donde el aporte de fluidos reproductivos en las IVP mejora los resultados asemejándose a aquellos in-vivo.

We aimed to describe such effects in pigs. Blood samples and growth parameters were collected from piglets derived from in vitro-produced embryos (IVP) with or without RF added in the culture media versus those artificially inseminated (AI), from day 0 to month 6 of life. An oral glucose tolerance test was performed on day 45 of life. We show here the first comparative data of the growth of animals produced through different assisted reproductive techniques, demonstrating differences between groups. Overall, there was a tendency to have a larger size at birth and faster growth in animals derived from in vitro fertilization and embryo culture versus AI, although this trend was diminished by the addition of RFs to the culture media. Similarly, small differences in hematological indices and glucose tolerance between animals derived from AI and those derived from IVP, with a sex-dependent effect, tended to fade in the presence of RF. The addition of RF to the culture media could contribute to minimizing the phenotypical differences between the in vitro-derived and AI offspring, particularly in males.

However, in pig, the combination of either procedures, or even their use separately, is still considered suboptimal due to the low efficiency of IVP plus the difficulty of performing ET in the long and contorted uterus of the sow. In addition, the potential impact of these two ART on the health of the offspring is unknown. We investigated here if the use of a modified IVP system, with natural reproductive fluids (RF) as supplements to the culture media, combined with a minimally invasive surgery to perform ET, affects the output of the own IVP system as well as the reproductive performance of the mother and placental molecular traits. Results: The blastocyst rates obtained by both in vitro systems, conventional (C-IVP) and modified (RF-IVP), were similar. Pregnancy and farrowing rates were also similar. However, when compared to in vivo control (artificial insemination, AI), litter sizes of both IVP groups were lower, while placental efficiency was higher in AI than in RF-IVP. Gene expression studies revealed aberrant expression levels for PEG3 and LUM in placental tissue for C-IVP group when compared to AI, but not for RF-IVP group. Conclusions: The use of reproductive fluids as additives for the culture media in pig IVP does not improve reproductive performance of recipient mothers but could mitigate the impact of artificial procedures in the offspring.
