    Somatostatin 14 affects the pituitary-ovarian axis in infant rats

    The effects of multiple somatostatin (SRIH- 14) treatment on the pituitary-ovarian axis were examined in infant rats. Female Wistar rats received subcutaneously two daily 20 µg/100g b.w. doses for five consecutive days (from 11 to 15 days of age). Changes in cell volume, volume density and number per unit area (mm2) of follicle-stimulating (FSH), luteinizing (LH) and somatotropic (GH) immunolabeled cells were evaluated by stereology and morphometry. Serum FSH and LH concentrations were determined by RIA. Ovaries were analyzed by simple point counting of follicles. SRIH-14 treatment significantly reduced FSH and LH cell volume, while their volume density and number per unit area were unaltered. Serum concentrations of FSH and LH were significantly reduced. Volume and volume density of GH cells was significantly decresed after SRIH-14 treatment, while their number per unit area was unaltered. In the ovary, SRIH-14 induced a significant increase in the percentage of primordial follicles followed by a significant decrease in percentage of primary follicles. The number of healthy and atretic preantral follicles was unchanged. It can be concluded that SRIH-14 treatment during the infantile period markedly inhibits pituitary FSH, LH and GH cells. In the ovary, SRIH-14 acts by inhibiting initial folliculogenesis without affecting atretic processes.

    Structural and ultrastructural modifications of adenohypophyseal gonadotropic cells in goat (Capra hircus)

    The structural and ultrastructural modifications of the gonadotropic cells of goats were studied with an immunohistochemical method (peroxidase-antiperoxidase), in anoestrus, gestation and milk production. The cell type which predominates in anoestms corresponds in its morphological characteristics to the classic FSH cells, and has two populations of homogeneous and electrodense secretory granules (141 - 244 nm and 244 - 400 nm in diameter), rough endoplasmic reticulum of flat cistemae and many largesized lysosomes. During gestation secretory granules show a characteristic reduction in size and are less abundant; lysosomes are also more scarce and the endoplasmic reticulum shows a high development; dilated and intercommunicated cisternae show a slight electrondense content, characteristic of typical LH cells. During milk production the cells show an increase in the number of secretory granules which are still small, and an increase in the number of lysosomes which appear as in anoestrus.

    The effects of cyclophosphamide on the gonadotrophic cells of the normal rat

    The effects of the administration of cyclophosphamide to male rats at doses of 400 mg/m2 for a period of 5 days and of 200 mg/m2 over two five-day cycles interrupted by a 21-day break reveal differences in the ultrastructural morphology of gonadotrophic cells between treated animals and normal controls. The ultrastructural data obtained coincide with the results obtained from the measurement of serum FSH and LH levels, indicating hypofunction. A significant fall was also recorded in blood testosterone levels. The two types of treatment did not give rise to morphological or functional differences.
