Abstract
Los estudios ultraestructurales en talos liquénicos se han incrementado en
los últimos años. La técnica de criofractura aborda la posibilidad de estudiar ultraestructuralmente material procedente de talos liquénicos sin necesidad de que éstos
sean sometidos a fijación química y a inclusión en resinas. Recientemente la técnica
de criofractura se ha convertido en una de las técnicas preparativas más importantes
para estudios ultraestructurales, fundamentalmente a nivel de membrana.
Aunque ésta técnica presenta un método físico independiente para elucidar
la ultraestructura liquénica, en este caso se ha completado el estudio de Lobaria pulmonaria con observaciones paralelas por los métodos convencionales de microscopía electrónica de transmisión.
Dado que la membrana del ficobionte parece estar implicada en su totalidad
en el proceso de transferencia de metabolitos, se ha hecho especial hincapié en la
observación de las partículas intramembranosas lMPs y de las invaginaciones del
plasmalema.
The ultrastructural studies on lichens have been increased during the last years. The freeze-etching technique deals with the possibility of studying ultrastructurally, material coming from lichen thallus without any need that these will be submited to a chemical fixation and to an inclusion on resins. Recently, the technique of freeze-etching has became one of the preparative techniques more important to ultrastructural studies fundamentally to a membrane level. Though, this technique presents a physical method independently to elucidate the ultrastructure of lichen, in this case, this study has been completed with parallel observations by the conventional methods of Transmission Electron MicrosCOPY. As the phycobiont's membrane seems to be involved in its totality in the process of transference of metabolites, an special remark on intramembrane particles (IMPs) and on the ridges of the plasmalemma, was made.
