- PublicationOpen AccessEffects of sediment sorbed linear alkylbenzene sulphonate on juveniles of the Senegal sole, Solea senegalensis, Toxicity and histological indicators(Murcia : F. Hernández, 2008) Hampel, M.; Ortiz-Delgado, J.B.; Sarasquete, C.; Blasco, J.Many synthetic organic substances, including surfactants, tend to be sorbed on suspended solids and to accumulate finally on bottom sediments, where benthic communities may be exposed to them. Concentrations of Linear Alkylbenzene Sulphonates (LAS) have been detected in estuarine and coastal sediments, presenting wide concentration ranges depending on the presence of treatment facilities, hydrodynamic conditions, organic matter content, etc. Senegal sole, Solea senegalensis, larvae (40 days posthatching; dph) were exposed to increasing concentrations of LAS spiked sediments, comprised between 0.37 and 880.78 mg LAS·kg-1 during 30 days. The obtained results showed that survival of exposed larvae was not significantly affected at environmentally relevant concentrations, the LC50 value being obtained after 30 days 876.46 mg·kg-1. However, the histological and histopathological analyses carried out in target organs revealed, that first alterations from the normal pattern were observed at concentrations of 222.66 mg·kg-1, presenting effects such as blood extravasation and hyperplasy of the lamellar epithelium in gills, increase of inter-myotomal spaces of the skeletal musculature and edematous separation of the skin from epidermis. At the highest exposure concentrations (755.27 and 880.78 mg LAS·kg-1), shrinkage of hepatocytes, nuclear pycnosis and blood stagnation are observed in the liver, degeneration of pancreatic cells, reduction of hemocytopoietic tissue in the kidney and vacuolisation of intestinal enterocytes was observed at histological level, as well as severe separation of the epidermis from the underlying tissues. Simultaneously, a significant increase of the wet weight with exposure concentration was observed in the test organisms.

Hernández, 1996) Sarasquete, C.; Gonzalez de Canales, M.L.; Arellano, J.M.; Muñoz-Cueto, J.A.; Ribeiro, L.; Dinis, M.T.Histochemical distribution of glycoproteins, carbohydrates and proteins rich in different aminoacids were studied using histological and histochemical procedures, in Senegal sole, Solea senegalensis (Kaup, 1858) larvae from hatching until day 15. Glycogen, proteins and glycoproteins were detected in the yolk-sac of the larvae at hatching and during the yolk-resorption. The epithelia1 digestive system (brush border, enterocytes and goblet cells) contained neutral and acid mucins (carboxylated andlor sulphated). Glycogen was observed in the cytoplasm of the digestive absortive cells (enterocytes) and in the liver (hepatocytes) on day 3-4 posthatching. Protein reactions, and specially those that showed proteins rich in arginine, tyrosine and tryptophan, were very intense in the zymogen granules of the pancreatic cells. Oesophageal and intestinal goblet cells contained glucose N-acetyl and sialic acid residues, but the mucin content of these mucous cells did not show affinity towards Con-A, suggesting the absence of glycoproteins with Mannose andlor glucose residues. WGA showed a very intense positivity in the microvilli of the digestive epithelium of the larvae and positive granules for both lectins, specially for Con-A, were detected in the cytoplasm of the anterior intestinal enterocytes.

Hernández, 1998) Sarasquete, C.; Gonzalez de Canales, M.L.; Arellano, J.M.; Muñoz Cueto, J.A.; Ribeiro, L.; Dinis, M.T.A battery of horseradish peroxidaseconjugated lectins (Con A, WGA and DBA), as well as conventional histochemical techniques (PAS, saponification, Alcian Blue pH 0.1, 1, 2.5, chlorhydric hydrolisis, neuraminidase, Bromophenol blue, Tioglycollate reduction and Ferric-ferricyanide-FeIII) were used to study the content and distribution of carbohydrates, proteins and glycoconjugate sugar residues on the skin and gills of Senegal sole, Solea senegalensis larvae and adults. During larval development of Solea senegalensis (from hatching until day 45 posthatching), epidermal sacciform, as well as branchial and epidermal chloride cells were unreactive with all cytochemical tests performed in this paper. Mucous or goblet cells of the corporal skin and gills containing strongly sulphated acid glycoproteins were evident on days 15-20 of larval development, as well as in epidermal and branchial mucous cells of adult specimens, which also contained GlcNAc andlor sialic acid. In adult specimen, the proteic content was higher in branchial mucous cells than in epidermal cells. In larvae, variable amounts of glycoproteins containing sialic acid, GlcNAc, GalNAc, Man andlor Glc residues were observed in epithelia1 cells and/or cuticle. GlcNAc andlor sialic acid sugar residues were only weakly detected in glycoproteins of some epidermal and branchial mucous cells of larvae by day 45, because from hatching until metamorphosis, lectin reactions (WGA, Con A and DBA) were negative in mucous cells.

Hernández, 2001) Arellano, J.M.; Storch, V.; Sarasquete, C.An histological and histochemical study was conducted on the stomach of adult Senegal sole, Solea senegalensis specimens. The stomach was made up of four distinct layers: mucosa, lamina propria-submucosa-, muscularis and serosa. Surface epithelial, glandular and rodlet cells were present in the mucosa. Cells of the columnar epithelium contained a basa1 nucleus. Numerous mitochondria, granular endoplasmic reticulum and Golgi apparatus consisting of severa1 parallel cisternae and vesicles were observed in the cytoplasm of these cells. The lysosomes were small, round and dense. The gastric glands were numerous in the pyloric and fundic regions but absent in the cardiac stomach. These glands were formed by two cell-types: light and dark cells. The light cells were characterised by numerous mitochondria, while dark cells had slightly fewer mitochondria and a tubulo-vesicular system. Rodlet cells similar to those observed in other teleostean fish were present among the epithelial cells. Although the epithelial cells of the mucosa contained a weak presence of neutral and acid mucopolysaccharides/mucosubstances, these substances were abundant in the lamina propria-submucosa. Proteins rich in arginine, lysine, cysteine and cystine were rarely present in the mucosa and lamina propriasubmucosa of stomach, while proteins rich in tyrosine were abundant in these layers. Acid phosphatase, and ATP-ase (pH 7.2 and 9.4) activities were detected in the mucosa and lamina propria-submucosa. Alkaline phosphatase activity was not detected.

Hernández, 2005) García-López, A.; Martínez Rodríguez, G.; Sarasquete, C.Despite the fact that the Senegalese sole Solea senegalensis is a target species for the aquaculture industry in Spain and Portugal, very little is known about its gametogenesis and especially about testicular development. Therefore, the male reproductive system in adult S. senegalensis was described using a histological and histochemical approach. Mean gonadosomatic index was very low (0.094±0.004%) and suffered slight changes throughout the experimental period. In transverse sections, the testis presents a reniform structure with two main regions, a cortical one with seminiferous lobules where germ cells proliferate in spermatocysts (germ /Sertoli cells units), and a medullar one with spermatic ducts that collect and store the produced sperm. The germinal compartment is organized into branching lobules of the unrestricted spermatogonial type, although the majority of type A spermatogonia are located at the distal part of the lobules. Spermatogenesis seems to be semi-cystic, since spermatids are released into the lobule lumen, where they are transformed into spermatozoa. Proteins in general, especially those rich in arginine, and carboxylated mucosubstances/glycoproteins increased from spermatogonia to spermatozoa. 3ß-Hydroxysteroid dehydrogenase enzymatic activity was exclusively observed in Leydig cells. The present study provides the first precise description of male reproductive apparatus in S. senegalensis.
