- PublicationOpen AccessDiversity patterns of benthic macrofauna caused by marine fish farming(Basilea: MDPI, 2011) Sanz Lázaro, Carlos; Marín Atucha, Arnaldo Aitor; Ecología e HidrologíaThis paper reviews the patterns observed in the diversity and structure of the macrofauna benthic community under the influence of fish farming. First, we explain the effects of organic enrichment on the sediment and the consequences for the inhabiting communities. We describe the diversity trends in spatial and temporal gradients affected by fish farming and compare them with those described by the Pearson and Rosenberg model. We found that in general terms, the trends of diversity and other community parameters followed the Pearson and Rosenberg model but they can vary to some extent due to sediment local characteristics or to secondary disturbances. We also show the different mechanisms by which wild fish can affect macrofauna diversity patterns under fish farming influence. In addition, we comment the importance of the macrofauna diversity in the ecosystem functions and propose some guidelines to measure functional diversity related to relevant processes at ecosystem level. We propose more research efforts in the main topics commented in this review to improve management strategies to guarantee a good status of the diversity and ecosystem functioning of sediments influenced by fish farming.

It can be used for bacterial infections in fish of all body sizes. However, physiological differences in fish depending on size may change the pharmacokinetics of danofloxacin and therefore its therapeutic efficacy. In this study, the change in the pharmacokinetics of danofloxacin in rainbow trout of various body sizes was revealed for the first time. The objective of this investigation was to compare the plasma and tissue pharmacokinetics of danofloxacin in rainbow trout of different body sizes. The study was conducted at 14 ± 0.5 ◦C in fish of small, medium, and large body size and danofloxacin was administered orally at a dose of 10 mg/kg. Concentrations of this antimicrobial in tissues and plasma were quantified by high performance liquid chromatography with ultraviolet detector. The plasma elimination half-life (t1/2Lz), volume of distribution (Vdarea/F), total clearance (CL/F), peak concentration (Cmax), and area under the plasma concentration–time curve (AUC0–last) were 27.42 h, 4.65 L/kg, 0.12 L/h/kg, 2.53 μg/mL, and 82.46 h·μg/mL, respectively. Plasma t1/2Lz, AUC0–last and Cmax increased concomitantly with trout growth, whereas CL/F and Vdarea/F decreased. Concentrations in liver, kidney, and muscle tissues were higher than in plasma. Cmax and AUC0–last were significantly higher in large sizes compared to small and medium sizes in all tissues. The scaling factor in small, medium, and large fish was 1.0 for bacteria with MIC thresholds of 0.57, 0.79, and 1.01 μg/mL, respectively. These results show that therapeutic efficacy increases with body size. However, since increases in danofloxacin concentration in tissues of large fish may affect withdrawal time, attention should be paid to the risk of tissue residue.

Fish were fed four different diets for 8 weeks: three groups of fish were fed diets containing 1%, 5% or 10% (w/w) fenugreek seeds, while the control group was fed a diet without fenugreek. At the end of the experiment, the expression gene of the main antioxidant enzymes (superoxide dismutase, catalase and glutathione reductase) was measured in the fish liver. Supplementation with fenugreek seeds at 10% significantly increased the growth of fish while different effects on gene expression were detected. An increase in H2O2 and OH⋅ scavenging ability and oxidative stability was observed in fish fed fenugreek-supplemented diets. In the present study, an 8-week supplementation of the diet of gilthead seabream specimens with fenugreek seeds resulted in an improvement of fish growth and antioxidant status (ROS scavenging, catalase and oil oxidative stability). These results afford a new perspective on the use of spices with medicinal properties as a supplement in fish feed in order to improve their growth and antioxidant status.
- PublicationOpen AccessEffects of cantharidin on fish erythrocytes, tumour cell lines, and marine pathogenic bacteriaCampos-Sánchez, José Carlos; Esteban Abad, María de los Ángeles; Guardiola Abellán, Francisco Antonio; Biología Celular e HistologíaCantharidin, a toxic monoterpene secreted by blister beetles, has long been used in traditional Chinese and modern medicine for its unique properties. However, despite its widespread use, its effects on fish have not been studied in depth. The aim of this study was to evaluate the potential therapeutic applications of cantharidin in fish by examining its antioxidant, hemoagglutinating, hemolytic and cytotoxic activities at different concentrations (0, 0.625, 1.25, 2.5, 5 and 10 μg mL-1) in three different cell lines. In addition, the study explored the bactericidal and bacteriostatic properties of cantharidin against various fish pathogenic bacteria. The results revealed that there were no significant differences in antioxidant, hemagglutinating or hemolytic activities between the different concentrations of cantharidin tested. However, the study found that cantharidin exhibited dose- and time-dependent cytotoxicity in sea bream (Sparus aurata) erythrocytes and in SAF-1, PLHC-1 and Hela cell lines, resulting in morphological changes indicative of apoptosis. Interestingly, the highest dose of cantharidin tested demonstrated potent bactericidal activity against four marine fish opportunistic bacteria, including Vibrio harveyi, V. anguillarum, Photobacterium damselae and Tenacibaculum maritimum, but no statistically significant changes in bacteriostatic activity were observed against any of the bacteria tested. Overall, these results provide valuable information on the potential therapeutic applications of cantharidin in fish aquaculture. Further research is needed to fully understand the mechanisms of action and to explore possible preventive uses of cantharidin in fish.

Antimicrobial peptides (AMPs) might constitute potential antiviral agents, which had been previously evaluated in fish with positive prospects, based on their properties as immunomodulators or directly killing pathogens. In this line, we aimed to evaluate this dual role by administering two European sea bass synthetic AMPs (Hamp or Dic) prior to NNV infection. Both treatments conferred partial protection against NNV though viral replication and load were not affected. Both AMPs elicited, prior to infection, AMP response and leukocyte mobilization whilst downregulated pro-inflammatory markers. Upon infection, Hamp and Dic peptides abrogated the inflammatory response provoked by NNV as well as avoid NNV-induced disturbance of the leucocyte distribution in the brain, mainly neutrophils, macrophages and CD8+ T cells. This study points that preventive applications of synthetic Hamp and Dic peptides exert their antiviral actions through the immunomodulatory role and not by a direct action of the antimicrobial on NNV. This work opens the door to the use of AMPs as potential prophylactic tools against NNV as well as immunostimulant in fish farms

Flesh, brain, liver and gonads were sampled for fatty acid analysis. Fish aggregated near sea-cages showed accumulation of fatty acids of vegetable origin in the studied tissues, due to surplus feed consumption or via predation of fish that consumed the feed. Gonads accumulated vegetable fatty acids in different manner in the different species, and the species least and most influenced by fish-feeds were selected for gonad histological examination. Results showed an acceleration of the final stages of the oocyte development in fish aggregated near fish farms compared to fish captured at long distance. Differences in oocyte development were more acute in the species which incorporated higher quantities of vegetable fatty acids.
- PublicationRestrictedMolecular and morphological identification of Cardicola (Trematoda: Aporocotylidae) eggs in hatchery-reared and migratory Atlantic bluefin tuna (Thunnus thynnus L.)(Elsevier, 2015-07-03) Forte-Gil, Débora; Holzer, Astrid S.; Pecková, Hana; Bartošová-Sojková, Pavla; Dolores, Emilio Ma.; Muñoz, Pilar; Peñalver García, José; Sanidad AnimalA microscopic and a molecular approach was used to investigate blood fluke infection in the very first specimens of Atlantic bluefin tuna (Thunnus thynnus, Scombridae) that were born in captivity and that were never moved from their specific cage culture sites in the Mediterranean. Data were compared with infection in wild, migratory bluefin tuna which were captured and fattened along the Spanish Mediterranean coast. Wild and cultured pop ulations sampled in the present study harbored the same Cardicola spp. richness and showed statistically insig nificant differences in overall parasite prevalence. Three morphotypes, crescent-shaped, fusiform and oval eggs, were observed in both non-migrant and migrant tuna. According to partial ITS2 and 28S rDNA sequences they belong to Cardicola opisthorchis, Cardicola forsteri, and to Cardicola sp., a recently described species, phyloge netically closely related to Cardicola orientalis. All three species were simultaneously present in some specimens. No significant difference in overall parasite prevalence was observed, though the prevalence of eggs of individual species, i.e. C. opisthorchis and C. forsteri, was significantly higher in fattened bluefin tuna. Our results strongly support a Mediterranean origin of the three Cardicola species encountered in cultured specimens and suggest that the intermediate hosts of these Cardicola species occur along the Mediterranean coast. Thus, infection with these parasites is not exclusively a result of infections happening along the migratory routes of wild bluefin tuna, in the Atlantic Ocean, as previously suggested. Once transferred to the sea for fattening, Atlantic bluefin tuna are at risk of infection with up to three different species of the genus Cardicola in along the Mediterranean coast in southeast Spain. On the contrary, C. orientalis infections might be associated to long-distance migration and an overlapping Thunnus maccoyii and Thunnus orientalis distribution. Statement of relevance This is the very first parasitological study conducted in T. thynnus born in captivity and cultured local in the Mediterranean. The results show that once transferred to the sea for fattening, they are at risk of infection with up to three different species of the genus Cardicola. These results strongly imply a Mediterranean distribution of these species.
