    Cardiovascular response to stress in patients with drug-resistant epilepsy: preliminary data

    El estrés es un factor de riesgo para las enfermedades cardiovasculares en la población general. La epilepsia se ha considerado un modelo adecuado de estrés crónico, con mayor incidencia de mortalidad cardiovascular que la población general. Este estudio proporciona datos preliminares sobre la respuesta cardiovascular ante un estresor cognitivo y una evaluación neuropsicológica en pacientes con epilepsia, explorando las relaciones entre la respuesta cardiovascular y el rendimiento cognitivo dependiendo del hemisferio responsable de la epilepsia. Treinta y cinco pacientes con epilepsia farmacorresistente, 17 con el área epileptógena(AE) en el hemisferio izquierdo (LH) y 18 con AE en el hemisferio derecho (RH), se sometieron a un estres cognitivo y una evaluación neuropsicológica. La respuesta cardiovascular se registró durante toda la sesión en ambas condiciones. Los resultados mostraron que una evaluación neuropsicológica de larga duración fue capaz de producir una respuesta cardiovascular modulada por el hemisferio, con disminuciones de la frecuencia cardíaca (y aumentos del intervalo RR) más pronunciadas en el período post-evaluación en pacientes con epilepsia del HI que en pacientes con epilepsia del HD (p = .05, p = .01, respectivamente). El hemisferio del AE moderó la relación entre la respuesta cardiovascular y el rendimiento cognitivo (para todos, p < .03). Nuestros hallazgos podrían tener implicaciones clínicas desde una perspectiva preventiva, ya que sugieren que el hemisferio del AE podría ser un factor relevante en el afrontamiento del estrés en personas con epilepsia farmacorresistente.

    Changes in Saliva Analytes Correlate with Horses’ Behavioural Reactions to An Acute Stressor: A Pilot Study

    Acute stress induces an array of behavioural reactions in horses that vary between individuals. Attempts to relate behavioural patterns and physiological responses have not always given clear-cut results. Here, we measured the changes in a panel of salivary components: salivary alpha-amylase (sAA), lipase, total esterase (TEA), butyrylcholinesterase (BChE), adenosine deaminase (ADA), and cortisol, and their potential link with horses’ behaviours after acute stress. Saliva samples were collected in nine riding horses subjected to a test consisting of opening an umbrella. Saliva sampling was obtained at a basal time point in the stall (T1), in the test indoor arena (T2), at a time of stress (T3), and 30 min (T4) and 60 min (T5) later. The horses’ behaviour was recorded at T3 for 1 min. sAA, lipase, TEA, and BChE showed significant changes along time, increasing at T3 for BChE, and decreasing at T4 for sAA and BChE. Butyrylcholinesterase appeared to be the most reliable predictor of behavioural responses, as it correlated with the index of emotionality, of laterality, and the occurrence of alarm signals, while sAA decreased when horses expressed quieter behaviours. These first results bring promising lines for novel, more precise physiological markers of acute stress in horses that can bridge the gap between behaviour and physiology.

    Daily rhythms of acute stress responses and antioxidant systems in the European sea bass (Dicentrarchus labrax): effects of the time of the year

    Fish reared in aquaculture face various acute stressors, including air exposure during handling. Research on the stress response in fish can provide essential insights into their physiology and help define better aquaculture practices. In this study, we investigated the daily rhythms in the stress-axis response of the European sea bass (Dicentrarchus labrax) subjected to an acute stressor consisting of air exposure (1 min), and how this response is influenced by the time of the day and the season of the year. In addition, rhythms in antioxidant systems were also assessed. The experiments were performed in late Autumn (December) and late Spring (June), with natural photoperiod (10 L:14D and 15 L:9D, respectively) and water temperature (ranging from 19.47 ± 0.17 °C in December to 22.13 ± 0.13 °C in June). Samples were collected throughout a 24-h cycle at Zeitgeber time (ZT) 0.5, 4, 7.5, 12, 16, 20, and 24.5 h at both seasons. At each sampling point, an untreated control (CTRL) group was sampled, while a STRESS group was exposed to air for one minute, returned to the tank, and sampled one hour later. Fish were sacrificed to collect plasma samples, hypothalamus and liver. Plasma samples were analyzed for cortisol, glucose, and lactate. In the hypothalamus, the mRNA expression levels of corticotropin-releasing hormone (crh) and crh-binding protein (crh-bp) were quantified using quantitative RT-PCR (qPCR). In the liver, genes related to antioxidant systems (catalase, superoxide dismutase 1, glutathione peroxidase,and glutathione reductase) and mitochondrial markers of stress (uncoupling protein 1, cytochrome c oxidase IV and peroxiredoxin3) were also analyzed by qPCR. The results revealed that most stress indicators (cortisol, cat, sod1, gsh-px, gsr, ucp1, coxIV) displayed daily rhythms. Furthermore, the stress response was significantly influenced by the time of day and the season in which the stressor was applied. In June, cortisol and glucose responses to stress were higher during the day than at night. The increase observed after stress in genes related to the antioxidant system was more significant in June than in December. Conversely, the response of mitochondrial markers was greater in December. Taken together, these findings highlight that the stress response of the European sea bass is time-dependent, both on a daily and a seasonal basis. This emphasizes the importance of considering cyclic environmental factors and circadian rhythms in aquaculture procedures to enhance fish welfare.

    Salivary testosterone measurements in growing pigs: validation of an automated chemiluminescent immunoassay and its possible use as an acute stress marker

    This study aimed to validate the use of an automated chemiluminescent immunoassay analyser for sal- ivary testosterone measurements in growing pigs and study how circadian pattern during daytime and stress can influence its values. The test method had intra- and inter-assay coefficient of variation lower than 10%. The method showed good linearity and recovery, and detection limits were low enough to detect salivary testosterone levels. No significant differences were observed in testosterone concentrations at different sampling time, and age and gender did not influence circadian pattern. In addition, this assay was used to quantify testosterone in two models of acute stress and, in both cases, significant increases (P < 0.01) in salivary testosterone were detected. Therefore, the automated assay system tested for porcine testosterone determinations would be suitable for its use in saliva samples and, furthermore, salivary testosterone levels could be used as a possible marker of acute stress in pigs.
